Effects of Resistin on Glucose Metabolism in C2C12 Myocytes

Results: To study the effects of resistin on glucose uptake ,glucose oxidation and glycogen synthesis in C2C12 myocytes,resistin cDNA was synthesizd byRT-PCR from mouse adipose tissue.resistin cDNA subcloned into PcDNA3.1(+) eukaryotic expression vector. The recombinant plasmids were transfected into C2C12 myocytes, the stable transfection cell line was screened by the culture medium containing G418. The expression of resistin protein of C2C12 myocytes was detected in by immunohistochemistry and Western blotting . C2C12 myocytes were divided into control, vector and resistin groups(n=6). According to the established method, detection of glucose uptake was performed with 3H labeled glucose, detection of glucose oxidation and glycogen synthesis were carried out with 14C-labeled glucose. ANOVA was used to evaluate the difference among groups. The analysis of the inserted DNA fragment nucleotide sequence was consistent with the resistin gene.The cells transfected by the recombinant plasmids can express resistin protein. Among three cell groups, there were significant difference in insulin stimulated glucose uptake, Glucose uptake rate in control, vector and resistin groups were7.81±0.87,7.88±1.50 and 5.79±1.26 respectively (P<0.05, n=6), while there were no significant difference about glucose uptake in basic state, glucose oxidation and glycogen synthesis in both of basic and insulin stimulated states.Resistin inhibited insulin mediated glucose uptake in C2C12 myocytes. Our findings may provide a clue to the role of resistin in the pathogenesis of IR in muscle.
